基于transformer2和doublesex級聯(lián)互作的煙粉虱性別決定機制研究

【摘要】:性別決定是生物兩性形成和正常生長發(fā)育的基礎,對種群延續(xù)意義重大。煙粉虱是一種世界性農業(yè)害蟲,為單雙倍體昆蟲。雄蟲來自未受精的卵為單倍體,雌蟲來自受精卵為雙倍體。為了研究煙粉虱的性別決定機制,對其性別決定中的兩個關鍵基因transformer2(Bttra2)和doublesex(Btdsx)進行了克隆和分析,并進一步利用RNAi技術闡述了其在煙粉虱性別決定中的作用。研究結果表明煙粉虱Bttra2全長1702bp,由10個外顯子和9個內含子組成;Btdsx全長2339bp,由7個外顯子和6個內含子組成。發(fā)現(xiàn)Bttra2有四種剪接形式,編碼四種蛋白,表達豐度雄蟲高于雌蟲;而Btdsx有13種剪接形式,編碼六種蛋白,表達豐度雄蟲也高于雌蟲。明確了transformer2和Btdsx在煙粉虱性別決定通路中均發(fā)揮至關重要作用且兩者相互影響和制約,Bttra2/Btdsx基因的干擾影響Vg基因和Btdsx/Bttra2的表達量,造成煙粉虱雄蟲生殖器官尾針畸形。研究結果對于闡明煙粉虱性別決定機制具有重要理論意義。
[Abstract]:Sex determination is the basis for the formation of biological hermaphroditism and normal growth and development, which is of great significance for population continuation. Bemisia Tabaci is a worldwide agricultural pest, a haploidy insect. Males from unfertilized eggs were haploid and females from fertilized eggs were diploidy. In order to study the sex determination mechanism of Bemisia Tabaci, two key genes, transformer2Bttra2) and DoublesexexBtdsx), were cloned and analyzed, and their roles in sex determination of Bemisia Tabaci were further elucidated by using RNAi technique. The results showed that the total length of Bemisia Tabaci Bttra2 was 1702 BP, which consisted of 10 exons and 9 introns, which consisted of 7 exons and 6 introns. It was found that Bttra2 had four splicing forms, encoding four proteins, and that Btdsx had 13 splicing forms, encoding six proteins, and expressed more abundant male than female. It is clear that transformer2 and Btdsx play an important role in the sex-determining pathway of Bemisia Tabaci and that the interference of Bttra2 / Btdsx gene affects the expression of Vg gene and Btdsx/Bttra2 and causes malformation of male reproductive organ of Bemisia Tabaci. The results are of great theoretical significance to clarify the sex determination mechanism of Bemisia Tabaci (Bemisia Tabaci).
