枯否細胞通過組織因子對大鼠急性胰腺炎凝血系統(tǒng)變化的影響

【摘要】:目的:觀察大鼠急性胰腺炎(acute pancreatitis,AP)組織因子(tissue factor,TF)及凝血系統(tǒng)的變化,探討枯否細胞(kupffer cells,KCs)通過組織因子對大鼠AP凝血系統(tǒng)的作用。方法:108只SD大鼠完全隨機分為假手術(shù)(sham operation,SO)組、輕癥胰腺炎(mild acute pancreatitis,MAP)組、重癥胰腺炎(severe acute pancreatitis,SAP)組、SO+氯化釓(Gd Cl3)組、MAP+Gd Cl3組、SAP+Gd Cl3組。Gd Cl3預(yù)處理組在誘發(fā)AP模型前24 h予尾靜脈注射Gd Cl3(10 mg/kg,0.1 ml),分別用1.5%和5%的牛黃膽酸鈉胰膽管逆行注射誘導(dǎo)MAP和SAP模型。建模后或假手術(shù)組分別于術(shù)后6、12、24 h處死大鼠,檢測其血清中淀粉酶及凝血項的變化;ELISA法檢測血清中TF的含量;分離并培養(yǎng)KCs,采用Western blot法檢測其TF的蛋白含量;免疫組化檢測肝臟中TF蛋白的表達;HE染色檢測胰腺組織病理損傷情況。結(jié)果:MAP和SAP組大鼠血清中淀粉酶含量明顯高于SO組(P0.01),與MAP+Gd Cl3組、SAP+Gd Cl3組各相應(yīng)時間點比較無明顯差異。同時間點MAP組和SAP組較SO組血清中TF含量明顯升高,MAP+Gd Cl3組和SAP+Gd Cl3組較MAP組和SAP組明顯降低(P0.05)。MAP和SAP組大鼠肝臟中TF蛋白表達較SO組表達明顯增加。MAP+Gd Cl3和SAP+Gd Cl3組KCs中TF蛋白含量較MAP組和SAP組明顯降低(P0.05)。SAP組較MAP和SO組大鼠血清凝血酶原時間(prothrombin time,PT)、活化部分凝血活酶時間(activated partial thromboplastin time,APTT)、纖維蛋白原濃度(fibrinogen,FIB)明顯升高,SAP+Gd Cl3組較SAP明顯降低(P0.05)。大鼠胰腺病理切片顯示,MAP組胰腺間質(zhì)水腫,SAP組胰腺呈凝固性壞死,伴有大量炎癥細胞浸潤,MAP+Gd Cl3和SAP+Gd Cl3組分別較MAP和SAP組病理損傷明顯減輕。結(jié)論:AP時KCs中TF表達明顯增加,并能導(dǎo)致凝血象的改變,抑制KCs功能可有效降低AP時TF的表達,減少凝血象的改變,從一定程度上可降低AP的發(fā)生和發(fā)展。
[Abstract]:Objective: To observe the changes of tissue factor (tissue factor, TF) and coagulation system of acute pancreatitis (AP) in rats and to explore the effect of Kupffer cell (Kupffer cells, KCs) on the coagulation system of rat AP through the tissue factor. Methods: 108 SD rats were completely divided into sham operation group and mild pancreatitis. Group Ancreatitis, MAP), group severe acute pancreatitis (SAP), SO+ gadolinium chloride (Gd Cl3) group, MAP+Gd Cl3 group, SAP+Gd group preconditioning group (10, 0.1, 0.1). The sham operation group died of 6,12,24 h after the operation to detect the changes of amylase and coagulation in serum, ELISA assay was used to detect the content of TF in serum; KCs was isolated and cultured, Western blot method was used to detect the protein content of TF; immunohistochemistry was used to detect the expression of TF protein in the liver; HE staining was used to detect the pathological damage of the pancreas. Results: MA The content of amylase in serum of P and SAP rats was significantly higher than that in group SO (P0.01). There was no significant difference in the corresponding time points between the MAP+Gd Cl3 group and the SAP+Gd Cl3 group. The content of TF in the MAP group and SAP group was obviously higher than that of the SO group. The expression of white expression in group SO was significantly higher than that in.MAP+Gd Cl3 and SAP+Gd Cl3 group, and the content of TF protein in group KCs was significantly lower than that of MAP and SAP groups (P0.05).SAP group than MAP and serum prothrombin time, activated partial thromboplastin time and fibrinogen concentration. FIB) significantly increased and SAP+Gd Cl3 group was significantly lower than SAP (P0.05). Pathological section of pancreas showed that pancreatic interstitial edema in group MAP, pancreatic necrosis in group SAP and infiltration of inflammatory cells in group SAP, MAP+Gd Cl3 and SAP+Gd Cl3 group were significantly less than MAP and other groups. Changes in coagulation and inhibition of KCs function can effectively reduce the expression of TF and reduce the changes of coagulation in AP, which can reduce the occurrence and development of AP to a certain extent.
