HIF-1-VEGF通路在SMVT大鼠腸道組織中的表達及意義

【摘要】:目的觀察HIF-1-VEGF通路信號分子在大鼠腸系膜上靜脈血栓形成(Superior mesenteric venous thrombosis,SMVT)腸道組織中的表達,探討該通路在SMVT中的作用。方法1 64只雄性SD大鼠,隨機分為兩組,假手術(shù)組(Sham組)和實驗組(SMVT組),每組大鼠32只。2 Sham組:僅打開腹腔,不阻斷血運,術(shù)后8h、24h、48h、72h分批處死大鼠;SMVT組:結(jié)扎法建立單純性周圍型SMVT模型,建模后8h、24h、48h、72h分批處死大鼠。3觀察腹腔及腸道大體變化;HE染色觀察大鼠腸道組織病理學(xué)變化;Real-time q PCR檢測大鼠腸道組織HIF-1α及VEGF m RNA的表達;免疫組織化學(xué)染色及Western blot檢測大鼠腸道組織HIF-1α及VEGF蛋白的表達。結(jié)果1大體觀察結(jié)果顯示:Sham組腹腔及腸道未見明顯變化;SMVT組大鼠腹腔出現(xiàn)不同程度及不同性狀積液,靜脈呈不同程度的怒張狀態(tài),腸道呈不同程度的腫脹及血液瘀滯狀態(tài),且均隨時間延長而逐漸減輕。2 HE染色結(jié)果顯示:Sham組大鼠腸道組織均無明顯病理變化;SMVT組大鼠腸道組織均出現(xiàn)不同程度紅細胞瘀滯,且隨時間延長瘀滯程度逐漸減輕。3 Real-time q PCR結(jié)果顯示:SMVT組較Sham組對應(yīng)各時間點HIF-1α及VEGF m RNA表達明顯升高(P0.05),且均呈遞減趨勢。4免疫組織化學(xué)染色及Western blot結(jié)果顯示:SMVT組較Sham組對應(yīng)各時間點HIF-1α及VEGF蛋白表達明顯升高(P0.05),且均呈先升后降趨勢。結(jié)論1 SMVT發(fā)病后,HIF-1α表達增加,并通過對其下游靶基因VEGF進行調(diào)控,啟動血管新生和生長過程,對缺血腸道組織側(cè)枝循環(huán)的建立具有重要意義。2SMVT發(fā)病后,VEGF在HIF-1α的調(diào)控下表達增加,促進血管的新生和生長,對缺血腸道組織側(cè)枝循環(huán)的建立具有促進作用。3 SMVT發(fā)病后,腸道組織缺血缺氧激活HIF-1-VEGF通路,信號分子HIF-1α及VEGF表達同步增加,促進血管新生和生長,加速側(cè)枝循環(huán)的建立,減輕血液瘀滯及微循環(huán)障礙,減少透壁性腸梗死的發(fā)生。
[Abstract]:Objective to investigate the expression of HIF-1-VEGF signaling molecule in the intestinal tissues of superior mesenteric venous thrombosis (SMBT) in rats, and to explore the role of this pathway in SMVT. Methods Sixty-four male Sprague-Dawley rats were randomly divided into two groups: sham-operated group (Sham group) and experimental group (32 rats in Sham group). The rats were killed in batches at 8 h, 24 h, 48 h and 72 h after operation. The simple peripheral SMVT model was established by ligation. Rats were sacrificed in batches at 8 h, 24 h, 48 h and 72 h to observe the gross changes of abdominal cavity and intestine. The histopathological changes of intestinal tissue were observed by HE staining. Real-time Q PCR was used to detect the expression of HIF-1 偽 and VEGF m RNA in rat intestinal tissue. Immunohistochemical staining and Western blot were used to detect the expression of HIF-1 偽 and VEGF protein in rat intestinal tissue. Results 1 the results of gross observation showed that there were no obvious changes in abdominal cavity and intestine of rats in the control group. In SMVT group, there were different degrees of effusion in abdominal cavity, different degree of irritation in vein, and different degree of swelling and blood stasis in intestinal tract of SMVT group. The results of HE staining showed that there were no obvious pathological changes in intestinal tissue of rats in the control group. All the intestinal tissues of SMVT group had different degrees of erythrocyte stasis. The results showed that the expression of HIF-1 偽 and VEGF m RNA in the Sham group was significantly higher than that in the Sham group, and the expression of HIF-1 偽 and VEGF m RNA in the Sham group was significantly higher than that in the Sham group. The expression of HIF-1 偽 and VEGF m RNA in the Sham group was significantly higher than that in the control group. 4. 4 immunohistochemical staining and Western blot showed that the HIF-1 偽 and VEGF m RNA expression in the Sham group were significantly higher than those in the control group. Compared with Sham group, the expression of HIF-1 偽 and VEGF protein increased significantly at each time point, and the expression of HIF-1 偽 and VEGF protein increased first and then decreased. Conclusion (1) the expression of HIF-1 偽 is increased after the onset of SMVT. By regulating the downstream target gene VEGF and initiating the process of angiogenesis and growth, it is important for the establishment of collateral circulation in ischemic intestinal tissue to increase the expression of SMVT under the control of HIF-1 偽.
