利用Identifiler分型系統(tǒng)推斷同胞關(guān)系

【摘要】:目的探討自主開發(fā)的同胞關(guān)系鑒定自動(dòng)分析軟件(ASI)對(duì)Identifiler分型系統(tǒng)進(jìn)行同胞關(guān)系鑒定的可行性。方法應(yīng)用本課題組所開發(fā)的軟件ASI,對(duì)151對(duì)同胞及31 224對(duì)人工模擬無(wú)關(guān)個(gè)體進(jìn)行Identifiler系統(tǒng)的15個(gè)常染色體STR基因座分型進(jìn)行分析,計(jì)算親權(quán)指數(shù)(PI)、同胞關(guān)系概率(WFS)和等位基因匹配情況,所獲數(shù)據(jù)進(jìn)行統(tǒng)計(jì)分析,自動(dòng)計(jì)算排序。結(jié)果當(dāng)WFS大于99.999%時(shí),同胞個(gè)體占39.07%,無(wú)關(guān)個(gè)體占0%,兩組具有顯著差異,可以推斷兩個(gè)體同胞關(guān)系。當(dāng)WFS介于1%~99.999%范圍內(nèi),同胞個(gè)體和無(wú)關(guān)個(gè)體有部分重疊,同胞個(gè)體占60.93%,無(wú)關(guān)個(gè)體占21.3%,兩者具有一定差異,可以通過(guò)增加檢測(cè)STR基因座,再結(jié)合案情加作Y-STR、m tDNA檢測(cè),以推斷兩個(gè)體是否具有同胞關(guān)系。當(dāng)WFS小于1%時(shí),同胞個(gè)體占0%,無(wú)關(guān)個(gè)體占78.7%,可以推斷兩個(gè)體不具有同胞關(guān)系。個(gè)體間等位基因匹配結(jié)果表明:在檢測(cè)Identifiler體系15個(gè)STR基因座時(shí),當(dāng)兩個(gè)體常染色體STR基因座的全相同數(shù)目≥5時(shí),或全不同數(shù)目≤1時(shí),提示為同胞關(guān)系;當(dāng)兩個(gè)體全不同數(shù)目≥6時(shí),或全相同數(shù)目≤1時(shí),提示為無(wú)關(guān)個(gè)體,以此作為預(yù)測(cè)有無(wú)同胞關(guān)系的界值。結(jié)論Identifiler系統(tǒng)及同胞關(guān)系鑒定自動(dòng)分析軟件ASI可用于推斷同胞關(guān)系。
[Abstract]:Objective to explore the feasibility of sibling relationship identification in Identifiler typing system developed by our own developed automatic sibling relationship analysis software (Identifiler). Methods using the software developed by our team, 151 pairs of sibling and 31 224 pairs of artificial simulation were tested without artificial simulation. The 15 autosomal STR loci of the Identifiler system were analyzed. The paternity index, sibling probability and allele matching were calculated. The obtained data were statistically analyzed and sorted automatically. Results when the WFS was greater than 99.999, the sibling individuals accounted for 39.07 and unrelated individuals took up 00.There was a significant difference between the two groups. It can be inferred that when WFS is within the range of 99.999%, there is a partial overlap between the sibling individual and the unrelated individual, the sibling individual occupies 60.9333um and the unrelated individual occupies 21.3B. there is a certain difference between the two. We can detect the STR locus by increasing the detection of the STR locus. Combined with the case, Y-STRM tDNA test is used to infer whether the two bodies have sibling relationship. When WFS is less than 1, It can be inferred that there is no sibling relationship between the two bodies. The results of allelic gene matching between individuals indicate that when 15 STR loci in the Identifiler system are detected, when the number of STR loci of two autosomes is equal to or greater than 5, the number of STR loci of two autosomes is more than 5.
