水稻和玉米葉肉細(xì)胞以及維管束鞘細(xì)胞轉(zhuǎn)錄組分析

  C4植物的光合作用效率高于C3途徑的植物接近50%。為了將C4途徑引入C3植物中,需要對(duì)調(diào)控光合作用的關(guān)鍵基因的分子作用機(jī)理進(jìn)行研究,特別是了解這些基因葉肉細(xì)胞(Mc)和維管束鞘細(xì)胞(BSc)中差異表達(dá)。因此,在本實(shí)驗(yàn)中的研究目的是:⑴從水稻和玉米中獲得均一Mc和BSc,以便通過(guò)利用LCM技術(shù)進(jìn)行細(xì)胞特定RNA測(cè)序;⑵分析水稻作為C3植物的特定Mc和BSc基因的差異表達(dá);⑶分析玉米作為C4植物的特定Mc和BSc基因的差異表達(dá)。水稻中已經(jīng)鑒定了包括Mc和BSc總共35956個(gè)基因,其中BSc特有2068個(gè),Mc特有5842個(gè)。參與光合作用的基因有:在Mc中特異性表達(dá)的OsSET13與OsSET26(Rubisco LS甲基轉(zhuǎn)移酶底物結(jié)合域)、CAB2R/OsLhcb1.2與RCABP89/OsLhcb2(葉綠素A-B結(jié)合蛋白)、PsaH與PsbX(光系統(tǒng)組分)、未知功能CHP01589、K09122、OsPM8ATP基因,但在BSc中這些基因也往往存在微量表...
