泡型肝包蟲病患者外周血Treg細胞的水平變化及其臨床意義

【摘要】:目的:通過流式細胞技術(shù)比較分析泡型肝包蟲病患者與健康人之間外周血CD4~+CD25~+CD127~(low)/Fox P3~+Treg細胞水平的差異,觀察他們之間的關(guān)系,探討其臨床意義。方法:選擇青海大學附屬醫(yī)院2016.8--2017.01期間住院患者25例作為實驗組,健康體檢者25例作為對照組。實驗組在術(shù)前和術(shù)后10天分別采集外周血標本,通過流式細胞技術(shù)比較分析泡型肝包蟲病患者與健康人之間外周血CD4~+CD25~+CD127~(low)/Fox P3~+Treg細胞水平的差異,分析他們之間的關(guān)系。結(jié)果:CD4~+CD25~+CD127~(low) Treg細胞水平:正常人(3.19±0.33)和患者術(shù)前(12.95±1.13)相比有統(tǒng)計學差異(P0.05),且患者術(shù)前的外周血CD4~+CD25~+CD127~(low)Treg細胞水平較高;正常人(3.19±0.33)和患者術(shù)后(5.70±1.21)相比有統(tǒng)計學差異(P0.05),且患者術(shù)后的外周血CD4~+CD25~+CD127~(low) Treg細胞水平較高;患者術(shù)前(12.95±1.13)和患者術(shù)后(5.70±1.21)相比有統(tǒng)計學差異(P0.05),且患者術(shù)前的外周血CD4~+CD25~+CD127~(low) Treg細胞水平較高。CD4~+CD25~+Fox P3~+Treg細胞水平:正常人(1.85±0.50)和患者術(shù)前(4.99±0.98)相比有統(tǒng)計學差異(P0.05),且患者術(shù)前的外周血CD4~+CD25~+Fox P3~+Treg細胞水平較高;正常人(1.85±0.50)和患者術(shù)后(2.84±0.57)相比有統(tǒng)計學差異(P0.05),且患者術(shù)后外周血CD4~+CD25~+Fox P3~+Treg細胞水平較高;患者術(shù)前(4.99±0.98)和患者術(shù)后(2.84±0.57)相比有統(tǒng)計學差異(P0.05),且患者術(shù)前的CD4~+CD25~+Fox P3~+Treg細胞水平較高。正常人的外周血CD4~+CD25~+CD127~(low) Treg細胞和CD4~+CD25~+Fox P3~+Treg細胞有相關(guān)性(r=0.568,P0.05),且兩指標呈正相關(guān)。結(jié)論:1.泡型包蟲患者CD4~+CD25~+CD127~(low)/Fox P3~+Treg細胞水平均顯著升高,可能與患者免疫抑制狀態(tài)形成有關(guān)。2.泡型包蟲病患者外周血CD4~+CD25~+CD127~(low)/Fox P3~+Treg細胞水平術(shù)前顯著高于手術(shù)后,提示根治性手術(shù)可能能夠影響患者體內(nèi)免疫狀態(tài)。3.據(jù)前兩點可知,外周血Treg細胞水平可在一定程度上反應治療效果、監(jiān)測病情復發(fā)。
[Abstract]:Objective: to compare and analyze CD4 ~ CD25 ~ CD127 of peripheral blood between patients with alveolar hepatic hydatid disease and healthy persons by flow cytometry. The difference of cell level of / / Fox P3 ~ Treg. Methods: 25 inpatients from the affiliated Hospital of Qinghai University from June 8 to January 2017.01 were selected as the experimental group. 25 healthy persons were used as control group. Peripheral blood samples were collected before and 10 days after operation in the experimental group. Comparative analysis of peripheral blood CD4 ~ CD25 ~ CD127 low between patients with alveolar hepatic hydatid disease and healthy persons by flow cytometry. The difference of cell level of / / Fox P3 ~ Treg. The relationship between them was analyzed. Results the Treg cell levels of CD4 ~ CD25 ~ CD127) in normal subjects were 3.19 鹵0.33) and those in patients before operation (3. 19 鹵0. 33). 12.95 鹵1.13) compared with the statistical difference (P0.05). The level of CD4 ~ CD25 ~ CD127~(low)Treg cells in peripheral blood was higher than that in patients before operation. There was significant difference between normal subjects (3.19 鹵0.33) and postoperative patients (5.70 鹵1.21) (P 0.05). The level of CD4 ~ CD25 ~ CD127 Treg cells in peripheral blood after operation was higher. There was significant difference between preoperative and postoperative patients (12.95 鹵1.13) and postoperative (5.70 鹵1.21) (P 0.05). The level of CD4 ~ CD25 ~ Fox P3 ~ Treg cells in peripheral blood was higher than that in patients before operation. There was significant difference between normal persons (1.85 鹵0.50) and postoperative patients (2.84 鹵0.57) (P 0.05). The level of CD4 ~ CD25 ~ Fox P3 ~ Treg cells in peripheral blood was higher after operation. There was significant difference between preoperative and postoperative patients (4.99 鹵0.98) and postoperative (2.84 鹵0.57) (P 0.05). The levels of CD4 ~ CD25 ~ Fox P3 ~ Treg cells were higher in patients before operation. Treg cells and CD4 ~ CD25 ~ Fox P3 ~ Treg cells were associated with rrmc0.568. P0.05). Conclusion the level of CD4 ~ CD25 ~ CD127~(low)/Fox P3 ~ Treg cells in patients with hydatid alveolar type increased significantly. 2. Peripheral blood CD4 ~ CD25 ~ CD127 of patients with alveolar hydatid disease may be related to the formation of immunosuppressive state in patients with hydatid disease. The cell level of / / Fox P3 ~ Treg was significantly higher before operation than that after operation. The results suggest that radical operation may affect the immune state of patients. According to the first two points, the level of peripheral blood Treg cells can respond to the therapeutic effect to a certain extent and monitor the recurrence of the disease.
