【摘要】：目的:研究微小RNA-7(miR-7)敲減(KD)對急性肝損傷(ALI)模型小鼠的影響。方法:野生型(WT)小鼠和miR-7KD小鼠腹腔注射30 mg/kg刀豆蛋白(ConA)建立急性肝損傷模型;48 h后,觀察小鼠肝臟的形態(tài)、重量及其臟器指數(shù)變化;HE染色觀察小鼠肝臟組織病理學(xué)變化;血清學(xué)方法檢查血清中谷丙轉(zhuǎn)氨酶(ALT)的水平;ELISA法檢測血清中細(xì)胞因子IL-4和IFN-γ的水平;流式細(xì)胞術(shù)檢測肝臟組織中CD4~+T細(xì)胞的比例及其相關(guān)的細(xì)胞因子IL-4和IFN-γ的表達(dá)變化。結(jié)果:與對照組相比,miR-7敲減后急性肝損傷小鼠的肝臟組織顏色變淺,重量減輕,重量指數(shù)明顯增加(P0.05);HE染色顯示miR-7KD小鼠血清炎癥細(xì)胞浸潤顯著增多;血清學(xué)方法檢測發(fā)現(xiàn)急性肝損傷小鼠血清ALT的水平明顯上升(P0.05);ELISA法檢測顯示miR-7KD小鼠血清中IFN-γ水平明顯升高(P0.01),而IL-4的表達(dá)水平則明顯降低(P0.01);流式細(xì)胞術(shù)檢測結(jié)果顯示,miR-7KD小鼠肝臟中CD4+T細(xì)胞比例顯著升高(P0.01),其相關(guān)的細(xì)胞因子IFN-γ的表達(dá)水平也顯著上調(diào)(P0.01),而IL-4的水平?jīng)]有發(fā)生明顯變化。結(jié)論:敲減miR-7基因可明顯促進(jìn)ConA誘導(dǎo)的小鼠急性肝損傷。
[Abstract]:Objective: to study the effect of minimal RNA-7miR-7 knockdown (KDD) on acute liver injury (ALI) mice. Methods: the acute liver injury model was established by intraperitoneal injection of concanavalin Cona for 30 mg/kg in wild-type mice and miR-7KD mice, and the morphology of liver was observed. The changes of weight and organ index were observed by HE staining, the levels of serum alanine aminotransferase (alt) and the levels of cytokines IL-4 and IFN- 緯 in serum were detected by serological method and Elisa respectively. The percentage of CD4T cells and the expression of related cytokines IL-4 and IFN- 緯 in liver tissue were detected by flow cytometry. Results: compared with the control group, the color and weight of liver tissue of mice with acute liver injury were lighter and lighter than those of control group. The weight index increased significantly (P 0.05) and HE staining showed that the infiltration of inflammatory cells in serum of miR-7KD mice was significantly increased. Serum levels of ALT in mice with acute liver injury were significantly increased by serological method. Elisa showed that the level of IFN- 緯 in serum of miR-7KD mice was significantly higher than that of P0.01G, while the expression of IL-4 was significantly decreased in mice with acute liver injury, and the results of flow cytometry showed that the level of IFN- 緯 in serum of miR-7KD mice was significantly higher than that of P0.01G, and that the expression of IL-4 was significantly decreased by flow cytometry. The proportion of CD4 T cells in the liver of miR-7KD mice was significantly increased, and the expression of IFN- 緯, the related cytokine, was also up-regulated, but the level of IL-4 did not change significantly. Conclusion: knockout of miR-7 gene can significantly promote the acute liver injury induced by ConA in mice.
