單一金納米電極的制備及應(yīng)用

【摘要】:近年來(lái),隨著對(duì)納米電極研究的深入,納米電極日益凸顯它的優(yōu)勢(shì),靈敏度高,響應(yīng)快,檢測(cè)限低,更小的RC常數(shù)以及更快的傳質(zhì)速率。因?yàn)槠湓陔姌O尺寸上的優(yōu)勢(shì),納米電極可應(yīng)用于研究電子傳遞動(dòng)力學(xué),檢測(cè)單個(gè)納米離子,作為探針用于SECM,生物活體及單細(xì)胞分析。眾所周知,金的表面由于具有更好的反應(yīng)活性使得金電極有更多的應(yīng)用,比如說(shuō)表面活性自組裝修飾,電催化,生物分析等等。在本文中,已經(jīng)制備半徑小于5nm的金納米盤(pán)電極以及單一金納米線電極,并且討論了其電化學(xué)行為,也研究了其在電化學(xué)生物傳感器方面的應(yīng)用。論文主要包括以下四個(gè)章節(jié):第一章:納米電極的概述,雙金屬核-殼結(jié)構(gòu),DNA簡(jiǎn)介,電化學(xué)DNA生物傳感器的研究進(jìn)展第二章:通過(guò)激光拉制的方法首次制備了半徑小于5nm的金納米盤(pán)電極,電化學(xué)表征,場(chǎng)發(fā)射透射電子顯微鏡(HR-TEM)和X-射線能譜分析法(EDX)對(duì)納米電極進(jìn)行了表征,電極表面修飾鐵蛋白的直接電化學(xué)研究了納米電極在電化學(xué)傳感方面的應(yīng)用。第三章:由金納米盤(pán)電極,通過(guò)氟化氫刻蝕技術(shù)制備成單一金納米線電極(Au-SPNEs)。再通過(guò)欠電位沉積(UPD)銅氧化置換技術(shù)制成鉑殼金核的金納米線電極(Au@Pt-SPNEs),PtCl42-置換UPD的銅層。場(chǎng)發(fā)射透射電子顯微鏡(HR-TEM)、X-射線能量射線譜分析法(EDX)和電化學(xué)的方法對(duì)納米線電極進(jìn)行了表征。實(shí)驗(yàn)表明我們制備的Au@Pt-SPNEs電極對(duì)甲醇、氧氣和雙氧水都有很好的催化效果。第四章:制備了基于金納米線電極(Au-SPNEs)的DNA生物傳感器。以亞甲基藍(lán)為電活性指示劑,采用方波伏安法進(jìn)行檢測(cè),該DNA生物傳感器很好的融合了“無(wú)試劑”污染和電化學(xué)的高靈敏響應(yīng)的優(yōu)點(diǎn)。這種新型的DNA傳感器將潛在的應(yīng)用于活體細(xì)胞中目標(biāo)分析物的實(shí)時(shí)檢測(cè)。
[Abstract]:In recent years, with the in-depth study of nano-electrode, nano-electrode increasingly highlights its advantages, such as high sensitivity, fast response, low detection limit, smaller RC constant and faster mass transfer rate. Because of its advantage in electrode size, nano-electrode can be used to study electron transfer kinetics, detect single nanoscale ion, and be used as probe for SECM, biological in vivo and single cell analysis. It is well known that the surface of gold has more applications due to its better reactive activity, such as surface active self-assembly modification, electrocatalysis, biological analysis and so on. In this paper, gold nanodisk electrode with radius less than 5nm and single gold nanowire electrode have been prepared. The electrochemical behavior of gold nanowire electrode and its application in electrochemical biosensor are also discussed. The thesis mainly includes four chapters as follows: chapter 1: Overview of nano-electrode, introduction of bimetallic core-shell structure DNA, Research Progress of Electrochemical DNA biosensor Chapter 2: gold nano-disk electrode with radius less than 5nm was prepared by laser pull method and characterized by electrochemistry. Field emission transmission electron microscopy (HR-TEM) and X-ray energy dispersive spectroscopy (edX) were used to characterize the nano-electrode. The application of nano-electrode in electrochemical sensing was studied by direct electrochemistry of ferritin modified on the electrode surface. Chapter 3: a single gold nanowire electrode (Au-SPNEs) was prepared by hydrogen fluoride etching. The au @ Pt-SPNEs / PtCl _ 42-replacement copper layer of au shell gold core was prepared by UPD copper oxidation replacement technique. The nanowire electrode was characterized by field emission transmission electron microscopy (HR-TEM) X-ray energy ray spectroscopy (edX) and electrochemical method. The experimental results show that the Autel-Pt-SPNEs electrode has good catalytic effect on methanol, oxygen and hydrogen peroxide. Chapter 4: a DNA biosensor based on Au-SPNEs was fabricated. Using methylene blue as the indicator of electrical activity and using square wave voltammetry, the DNA biosensor combines the advantages of "no reagent" pollution and high sensitivity of electrochemistry. This new type of DNA sensor will potentially be used for real-time detection of target analytes in living cells.
