丙泊酚脂肪乳注射液處方工藝優(yōu)化及質(zhì)量評價

【摘要】:丙泊酚(propofol)是一種靜脈注射用麻醉藥,由于其作用快、易控制等特點,其在臨床的應(yīng)用非常廣泛,本課題選用丙泊酚為主藥通過現(xiàn)代化的制備工藝,對丙泊酚脂肪乳注射液進行研究。建立了丙泊酚和有關(guān)物質(zhì)的高效液相色譜檢測方法,該方法靈敏度高、專屬性好,適用于丙泊酚及有關(guān)物質(zhì)的分析檢測,采用十八烷基硅烷鍵合硅膠作為填充劑,柱溫控制在30℃;分別采用275、254nm作為紫外檢測波長。流動相A為磷酸二氫鈉0.2%的水溶液,pH值為3.0,流動相B:乙腈,采用梯度程序洗脫法,流速為1.0 ml/min。在上述擬定的色譜條件下,有關(guān)物質(zhì)A和B均能與主峰丙泊酚得到良好分離,且相鄰峰之間的分離度超過1.5。同時采用蒸發(fā)光作為檢測器,建立了溶血磷脂酰膽堿的高效液相色譜檢測方法,該方法也具有靈敏度、專屬性好的特點。通過單因素試驗選擇了 1.2%磷脂和0.05%油酸用量,選擇了大豆油和中鏈甘油三酯比例為1:1的混合油作為油相載體。通過正交實驗設(shè)計來優(yōu)化制備工藝,最終選擇高剪切乳化溫度控制在70℃;NaOH選擇在初乳制備前加入,高壓均質(zhì)壓力為300bar,均質(zhì)溫度控制在60℃等工藝條件制備了丙泊酚脂肪乳注射液,并通過電子射鏡和粒徑分布等進行表征。在實驗室處方和制備工藝基礎(chǔ)上進一步進行中試工藝的優(yōu)化,使用1OObar壓力均質(zhì)替代實驗室的高剪切和均質(zhì)混合來進行初乳的制備,選擇了過濾壓力≤40Kpa,以減少大于1μm的不溶性微粒數(shù)量;灌裝充氮壓力為30Kpa時,其灌裝后的氧氣殘余量為3.0±0.4%;滅菌條件為121℃,12min,其中當工業(yè)蒸汽壓力≥0.3Mpa,冷卻水壓力=0.2Mpa時,在保證F0值≥12的條件下既能縮短升溫和降溫時間又能得到較低的破損率。采用優(yōu)化的中試生產(chǎn)工藝制得了丙泊酚脂肪乳注射液,其平均粒徑為245.2±10nm,其它各項質(zhì)量指標均符合要求。對丙泊酚脂肪乳注射液中試工藝生產(chǎn)的樣品進行了影響因素試驗、加速試驗和長期試驗,測定了 pH值、滲透壓摩爾濃度、丙泊酚含量、有關(guān)物質(zhì)、粒徑分布和溶血磷脂酰膽堿等,結(jié)果表明中試工藝制備的丙泊酚脂肪乳注射液在強光照射、低溫循環(huán)條件下穩(wěn)定,溶血磷脂酰膽堿對溫度比較敏感,6個月加速試驗和24個月長期試驗表明丙泊酚脂肪乳注射液各項質(zhì)量指標符合要求。
