大麥半矮稈基因sdw1/denso的克隆與功能分析

半矮稈基因廣泛用于大麥育種。迄今,已報導(dǎo)過30多種大麥矮稈基因。其中sdw1/denso半矮稈基因在歐美、澳大利亞等國家大量應(yīng)用,是大麥育種中應(yīng)用最成功的基因之一。而我國大麥矮稈育種上應(yīng)用的主要基因是uzul基因。sdw1/denso基因在不同環(huán)境條件下能使株高降低10-20 cm。此外,該基因與遲抽穗、晚熟、降低千粒重和高過篩率相關(guān)。sdw1/denso基因?qū)Ξa(chǎn)量的影響因遺傳背景而異;趕dw1/denso基因適宜的株高和潛在的增產(chǎn)效應(yīng),故已被轉(zhuǎn)入至許多大麥品種。目前,至少有4個sdw1/denso等位基因被先后報導(dǎo)。值得關(guān)注的是,denso基因主要用于啤酒大麥育種,而sdwl基因主要用于飼料大麥育種。綜上所述,有必要克隆sdw1/denso基因,以闡明其矮稈機(jī)制,揭示該基因?qū)r(nóng)藝性狀的影響,從而更好地應(yīng)用于大麥育種,F(xiàn)將本文的主要研究及取得的結(jié)果介紹如下:1.大麥sdw1/denso候選基因的克隆比較基因組學(xué)表明,大麥sdw1/denso基因位于與水稻1號染色體sdl基因染色體存在共線性區(qū)域,而水稻sdl基因編碼GA 20-氧化酶。通過PCR法在大麥中獲得部分水稻sdl基因的同源...
