基于復制-轉(zhuǎn)錄碰撞的突變和進化意義(英文)

復制和轉(zhuǎn)錄機器會同時使用相同的DNA區(qū)域作為模板,因此復制和轉(zhuǎn)錄不可避免地以頭對頭或追尾方式相互碰撞.頭對頭碰撞和追尾碰撞均會導致復制機器停留,從而造成DNA損傷和基因組不穩(wěn)定.就基因組完整性而言,頭對頭碰撞比追尾碰撞的后果更嚴重.本文回顧總結(jié)了復制-轉(zhuǎn)錄沖突的解決機制和進化影響.相對于前導鏈,滯后鏈上非同義(氨基酸改變)突變的發(fā)生率更高,并且滯后鏈上基因的高頻誘變?nèi)Q于轉(zhuǎn)錄本和基因大小,因此較快的適應(yīng)性突變發(fā)生在滯后鏈上.頭對頭基因的高度轉(zhuǎn)錄增加了復制過程中響應(yīng)壓力的突變率.無論是頭對頭還是追尾模式,復制-轉(zhuǎn)錄沖突都可能是適應(yīng)性進化的驅(qū)動力.
