原兒茶酸在雞組織中的殘留消除規(guī)律

【摘要】:為研究原兒茶酸在雞組織中殘留消除規(guī)律,利用高效液相色譜法(HPLC)在260nm處檢測原兒茶酸在雞組織中殘留情況,確定流動相體積比為乙腈∶水=9∶91,用H_3PO_4調(diào)pH至2.5。結(jié)果表明:試驗測得雞肝、腎、肌肉、皮膚和脂肪中檢測限分別為13.5、25.0、12.0和16.0ng/g,定量限為45、85、40和50ng/g;平均加標回收率為63.30%~102.65%,變異系數(shù)為1.01%~11.43%。以臨床推薦劑量20mg/kg(體重)口服給藥,每天2次,連續(xù)5d,腎臟、肝臟中原兒茶酸的殘留量最高,皮膚和脂肪、肌肉中殘留量較低。休藥4h后,各組織中原兒茶酸殘留量均低于最高殘留限量(肝臟1.52mg/g,腎臟3.05mg/g,肌肉0.51mg/g,皮膚和脂肪3.05mg/g)。試驗結(jié)果表明,原兒茶酸在雞組織中的殘留消除較快,新藥休藥期的制定應根據(jù)具體情況進一步研究確定,目前初步建議休藥期為0h。
